چکیده

The levels of ribonucleotide reductase activity (measured as the conversion of 3H-cytosine monophosphate to 3HdCMP) in cell-free extracts of partially synchronized cultures of L cells were measured as the cells moved through the division cycle. A strict parallelism was found between the measured enzyme levels and the fractions of the population that were forming DNA. This relationship between enzyme level and ability to replicate DNA indicated that the reductase activity fell rapidly when cells emerged from the DNA synthetic period, suggesting a lability of the enzyme in vivo. The lability in vivo was confirmed by measuring enzyme levels in cultures where further protein synthesis had been blocked with cycloheximide. Under these conditions, an apparent half-life of less than 2 hours was observed for ribonucleotide reductase in the L cell.

Ribonucleotide reductase M1 subunit in cellular proliferation, quiescence, and differentiation.

Ribonucleotide reductase catalyzes the first unique, rate-limiting step in DNA synthesis; both its large (M1) and small (M2) subunits are necessary for activity. While direct studies of M2 expression have previously shown a tight correlation with S phase, the kinetic features of M1-expressing cells have remained ill defined. Therefore we have, using immunofluorescence flow cytometry, analyzed c...

Liz1p, a novel fission yeast membrane protein, is required for normal cell division when ribonucleotide reductase is inhibited.

Ribonucleotide reductase activity is required for generating deoxyribonucleotides for DNA replication. Schizosaccharomyces pombe cells lacking ribonucleotide reductase activity arrest during S phase of the cell cycle. In a screen for hydroxyurea-sensitive mutants in S. pombe, we have identified a gene, liz1(+), which when mutated reveals an additional, previously undescribed role for ribonucleo...

Ribonucleotide reductase and cell proliferation. I. Variations of ribonucleotide reductase activity with tumor growth rate in a series of rat hepatomas.

The relationship between ribonucleotide reductase and tumor growth rate was measured in a series of rat hepatomas of different growth rates. An excellent correlation was found between tumor growth rate and the specific activity of ribonucleotide reductase. Differences of ZOO-fold in enzyme specific activities exist between the very fast and the slow growing hepatomas. This represents the larges...

Escherichia coli ribonucleotide reductase expression is cell cycle regulated.

The expression of the genes encoding ribonucleotide reductase in Escherichia coli was investigated in cultures synchronized by obtaining the smallest cells in a population after sucrose gradient centrifugation. Specific activity of ribonucleotide reductase and DNA initiation were found to increase in parallel, periodically as a function of the cell cycle. The expression of nrd was also determin...
